ABSTRACT

ObjectiveTo investigate the effect of Dendrobii Caulis mixture on cell inflammatory response and apoptosis in diabetic rat with non-alcoholic fatty liver disease(NAFLD) and its possible mechanism. MethodForty male SD rats were randomly divided into blank group and model group of type 2 diabetes mellitus(T2DM) with NAFLD according to body weight. The model was established by high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin(STZ), which was randomly divided into the model group, Dendrobii Caulis mixture group(16.67 g·kg-1·d-1) and metformin group(100 mg·kg-1·d-1) according to blood glucose and body weight, and 10 rats in each group. Rats in each group were administered by continuous gavage for 4 weeks, the blank and model groups were given saline by gavage at 10 mL·kg-1·d-1. Fasting blood glucose(FBG), serum insulin(INS), glycosylated serum protein(GSP), triglyceride(TG), total cholesterol(TC), high density lipoprotein(HDL-C), low density lipoprotein(LDL-C), alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected in each group of rats. The liver tissues of rats in each group were stained with hematoxylin-eosin(HE) to observe the pathological changes, and the positive expressions of nuclear transcription factor(NF)-κB, NOD-like receptor heat protein structural domain-related protein 3(NLRP3), interleukin(IL)-1β and tumor necrosis factor(TNF)-α were observed by immunohistochemistry. Western blot and fluorescence quantitative polymerase chain reaction(Real-time PCR) were used to detect the protein and mRNA expression of B-cell lymphoma-2(Bcl-2), Bcl-2 associated X protein(Bax), Caspase-3 and NF-κB p65, NLRP3, IL-1β, TNF-α in liver tissue of rats in each group. ResultCompared with the blank group, FBG, GSP, TC, TG, LDL-C, AST and ALT levels were increased, INS and HDL-C levels were decreased, Bax, Caspase-3, NLRP3, IL-1β, TNF-α protein and mRNA expression were increased in model group, the ratio of p-NF-κB/NF-κB protein increased, the expression of Bcl-2 protein and mRNA decreased, and the positive immunohistochemical expression of NF-κB, NLRP3, IL-1β and TNF-α increased, and the differences were statistically significant(P<0.01). The morphological structure of the liver was disrupted, and obvious fat vacuoles were seen. Compared with the model group, FBG, GSP, TC, TG, LDL-C, AST and ALT levels of Dendrobii Caulis mixture group and metformin group were decreased, INS and HDL-C levels were increased, and protein and mRNA expressions of Bax, Caspase-3, NLRP3, IL-1β and TNF-α were decreased, the protein ratio of p-NF-κB/NF-κB decreased, the expression of Bcl-2 protein and mRNA increased, and the positive immunohistochemical expressions of NF-κB, NLRP3, IL-1β and TNF-α decreased, and the differences were statistically significant(P<0.01). The liver morphology and structure were relatively complete, and the fat vacuoles were reduced. ConclusionDendrobii Caulis mixture can inhibit cell apoptosis, reduce inflammatory response and alleviate liver injury in rats with T2DM and NAFLD, the mechanism may be related to inhibiting the activation of NF-κB pathway, blocking the activation of NLRP3 inflammatory vesicles, reducing IL-1β secretion, attenuating Caspase-3 activity and reducing the ratio of Bcl-2/Bax.

ABSTRACT

Through consulting the ancient herbal and medical books, combined with the field investigation, the name, origin, collection and processing of Dendrobium medicinal materials were researched, which provided a basis for the development of famous classical formulas containing this kind of herbs. Due to the wide distribution of D. officinale, the Dendrobium species represented by D. officinale and D. huoshanense, which are short, fleshy and rich in mucus, should be the most mainstream of Dendrobium medicinal materials in previous dynasties. Compared with Shihu, Muhu with loose texture, long and hollow is born on trees. According to the characteristic description, it should be D. nobile, D. fimbriatum and so on, of which D. nobile was the mainstream. The Chinese meaning of Jinchai was confused in the past dynasties, so it was not suitable to be treated as a plant name. The production areas of Dendrobium medicinal materials in the past dynasties have changed with the discovery of varieties, artificial cultivation and other factors. Lu'an, Anhui province, was the earliest recorded in the Han and Wei dynasties. Since the Tang and Song dynasties, it had been extended to Guangdong and Guangxi, and it was considered that "Dendrobii Caulis in Guangnan was the best". In the Ming dynasty, Sichuan and Zhejiang products were highly praised, and in the Qing dynasty, Huoshan products were highly praised. Dendrobium medicinal materials had been used as medicine by stems in all dynasties. The medicinal materials were divided into fresh products and dry products. The fresh products can be used immediately after removing the sediment from the roots. The dry products need further processing, most of them used wine as auxiliary materials for steaming, simmer to paste or decoction into medicine. D. officinale and D. huoshanense have special processing specifications since the middle of Qing dynasty, that is, "Fengdou". According to the research results, in Ganluyin, the effect of Dendrobium medicinal materials is mainly heat clearing, and D. nobile with bitter taste can be selected. The main effect of Dendrobium medicinal materials in Dihuang Yinzi is tonic, D. officinale or D. huoshanense can be selected.

ABSTRACT

Life style changes with economic and social development, including a improper diet, cause diabetes, dyslipidemia, hypertension and other metabolic disturbances, which has become a major threat to the health of people around the world. Dendrobii Caulis, a traditional Chinese herbal medicine with various medicinal and health-care effects, is derived from different species of Dendrobium, which is traditionally used for “consumptive thirst syndrom”. In recent years, its preventive effects on metabolic disturbances have been attracting more and more attention with the high incidence of metabolic disturbances. This paper aims to review the recent achievements of different Dendrobium species in preventive and therapeutic effects and its traditional functions on metabolic disturbances, mainly in diabetes, hypertension and dyslipidemia. Furthermore, probable mechanisms were summarized based on its traditional functions. Dendrobii Caulis was effective on metabolic disturbances primarily relevant to its conventional functions including replenishing yin for maintaining gastric tonicity, promoting body fluid production, nourishing yin and clearing heat, which is helpful for treatment of spleen impediment with abdominal flatulence and interior heat, the source of various metabolic disturbances in traditional medicine. This review provids reference for mechanism research and clinical application of Dendrobium in metabolic disturbances prevention and treatment.

ABSTRACT

Dendrobium represents one of the most important orchid genera with ornamental and medicinal values. The stems of Dendrobium species have been used as Dendrobii Caulis in traditional Chinese medicine for a thousand years, which manifest a diversity of medicinal effects including tonic, analgesic, antipyretic, and anti-inflammatory. In recent decades, about 100 compounds have been found and identified from the medicinal parts of more than 40 Dendrobium species, which include polysaccharide, alkaloids, flavonoids, phenols, terpenoids, amino acids, trace elements, coumarin, tanning, sterols, etc. This review focus on main research conducted during the last decade on Dendrobii Caulis and their constituents, which have been studied on their pharmacological effects involving immunomodulatory activities, antidiabetic, anticancer, anti-oxidant, anti-aging, hepatoprotective, anti-inflammatory, neuroprotective, cardiovascular protective, enterogastric strengthening, anti-angiogenesis, allergic dermatitis symptoms improving, antifatigue, antiplatelet aggregation, etc. Simultaneously, the key research problems and the future development on Dendrobium are prospected.

ABSTRACT

Dendrobii Caulis was traditionally used for promoting middle energizer, strengthening enterogastric function to light body weight, and prolonging life according to Shennong's Classic of Materia Medica and many other previous ancient books of traditional Chinese medicine. The stomach and intestine tonic effect is still approved in modern society and broadly applied in clinic, whose efficacy has been featured as replenishing yin for maintaining gastric tonicity, enhancing the production of body fluid, and clearing away heat in Chinese Pharmacopoeia 2015. This review firstly made a summary of ancient literature on strengthening enterogastric function of Dendrobii Caulis, combined with literature of modern pharmacological research and clinical application from literature search system, such as CNKI, etc. It was summarized that Dendrobii Caulis had broad bioactivities in enterogastric system including gastric mucosa protection, digest-promotion, intestinal flora modulation, intestinal immunity regulation, stimulation of digestive juice secretion, etc. Furthermore, present situation of product development was analyzed based on patent and health product search system. This review could provide a reference for further development and clinical application of Dendrobii Caulis.
